Differential Expression

Compare Gene Expression Between Different Conditions

Differential expression analysis identifies variations in the expression of one or more genes between different biological or experimental conditions. It involves measuring the amount of RNA associated with each gene and comparing these expression levels across multiple samples.

This approach is widely used to understand how an organism reacts to a change in condition: exposure to a molecule, environmental modification, difference between two tissues, or comparison between several organisms.

The study of these variations notably highlights genes whose activity increases or decreases in a given situation.

Why Analyze Differential Expression?

Comparing gene expression levels helps to better understand the biological mechanisms involved in a studied phenomenon.

Differential expression analysis is notably used to:

  • identify genes involved in a specific biological response
  • study the impact of a treatment or an environmental factor
  • compare organisms or strains cultured under the same conditions
  • analyze differences between tissues, developmental stages, or physiological states
  • identify genes potentially associated with a particular biological process.

This approach is an essential tool for exploring the molecular mechanisms underlying a biological phenomenon.

Techniques Used

Several methods can be employed to analyze differential gene expression.

RT-qPCR

Real-time quantitative PCR (RT-qPCR) precisely measures the amount of cDNA corresponding to a gene of interest. This technique monitors the amplification of a genetic sequence during PCR cycles using a fluorescent signal proportional to the amount of amplified DNA.

It notably allows for:

  • comparing gene expression between several samples
  • precisely measuring expression variations
  • analyzing multiple genes in the same study.

Transcriptomic Sequencing (RNA-Seq)

High-throughput sequencing technologies allow for the analysis of the expression of thousands of genes simultaneously. Expression is estimated by counting RNA fragments corresponding to each gene in the studied samples.

This approach offers a global view of an organism's genetic activity.

Digital PCR (dPCR)

Digital PCR enables absolute quantification of nucleic acids, without a standard curve or reference sample. The sample is divided into several thousand individual and independent reactions, which provides high tolerance to inhibitors, superior precision, increased sensitivity, and excellent reproducibility.

It is particularly suitable for:

  • determining the copy number of a given DNA target
  • detecting rare mutations
  • measuring the viral load of a sample
  • accurately analyzing gene expression differences
  • quantifying a high-throughput sequencing library

Biomnigène is equipped with the QuantStudio Absolute Q plate digital PCR system.

Interpretation of Results

Differential expression analysis identifies genes referred to as differentially expressed, meaning their expression level varies significantly between two or more conditions.

These genes can be:

  • overexpressed in a given condition
  • underexpressed compared to a reference condition.

Identifying these variations provides valuable information for understanding the biological mechanisms involved in an organism's response.

An Analysis Tailored to Your Project

The study of differential expression can be applied to numerous scientific fields:

  • fundamental research
  • cell and molecular biology
  • microbiology
  • biotechnologies
  • medical research.

Depending on the nature of the samples and the objectives of the study, different analytical strategies can be implemented to obtain the most relevant results.
